Fig: Fluorescence immunohistochemical analysis of Rat-ileum tissue (Formalin/PFA-fixed paraffin-embedded sections). with Mouse anti-MUC2 antibody (AWA02739) at 1/400 dilution. The immunostaining was performed with the TSA Immuno-staining Kit (ABIOWELL, AWI0689). The section was pre-treated using heat mediated antigen retrieval with EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (AWA02739) at 1/400 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system followed by a separate fluorescent tyramide signal amplification system (red). DAPI (blue, AWC0291) was used as a nuclear counter stain. Image acquisition was performed with Slide Scanner. |

Fig: Immunocytochemistry analysis of HT-29 cells labeling MUC2 with Mouse anti- MUC2 antibody(AWA02739)at 1/100 dilution(Green ). Cells were fixed in 4% paraformaldehyde for 10 minutes at 37 ℃, permeabilized with 0.03% Triton X-100 in PBS for 30 minutes, and then blocked with 5% BSA for 60 minutes at 37 ℃. Cells were then incubated with Mouse anti- MUC2 antibody (AWA02739)at 1/100 dilution in 2% negative goat serum overnight at 4 ℃. Goat Anti-Mouse IgG H&L (iFluor™ 488, AWS0003) was used as the secondary antibody at 1/200 dilution for 60 minutes at 37 ℃. Nuclear DNA was labelled in blue with DAPI(AWC0291). |

The tissues were blocked in 3% H2O2 for 15 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (AWA02739) at 1/200 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system(ABIOWELL, AWI0629). DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX. |
